Barleria lupulina Lindl is a popular medicinal plant distributed in the mountains of South and West India. In the present work the effect of methanolic extract of aerial parts of B. lupulina on CNS activity has been evaluated. CNS activity was assayed in several experimental models, in mice and rats: general behavior, exploratory behavior, muscle relaxant activity, conditioned avoidance response, and sodium phenobarbital induced sleep time trials. The aerial parts of the B. lupulina plant were extracted with methanol and the solvent was removed by vacuum distillation. Methanol extract (100, 200 and 300 mg / kg) showed a reduction in the pattern of general behavior (spontaneous activity, alertness, awareness, pain response and touch response) in a dose-dependent manner. It was found that the extract produced a significant reduction of the exploratory behavior profile (labyrinth Y test, head immersion test) and conditioned avoidance response with all doses tested. The methanolic extract showed significant motor in coordination and muscle relaxant activity. The extract also potentiated the sleep time induced by sodium phenobarbital. Preliminary research showed that the methanol extract of B. lupulina has significant psycho-pharmacological activity.
